Those are ambiguous in themselves.
Why would you be addressing your sister or your colleague so formally in the first place instead of by their names.

Therefore:
"This is Jane, my sister."
to which your sister replies,
"Thank you, my brother. Pleased to meet you, Jane."

If you are going to provide examples, they should be simpler, my TC colleague. (Did you see that? Well did you?) :razz:

My Spanish grammar book comes in handy too.
